Depo-Provera is one of the most widely used forms of hormonal birth control.

Less than a year later, another study confirmed the original paper’s findings. Researchers now believe that women who take Depo-Provera are up to 5.5 times more likely to be diagnosed with meningiomas than women who have never taken this medication.

Pfizer, however, did not update its labeling for Depo-Provera until December 2025.

As a result, most women who take or have taken Depo-Provera were never warned that their contraceptives could cause brain tumors.

The Science Behind Depo-Provera

Depo-Provera is the brand name of an injectable contraceptive made by Pfizer.

Unlike common types of oral birth control, Depo-Provera is administered once every three months as a shot. Depo-Provera works by altering hormone levels in the body, suppressing ovulation, and triggering mucus production around the cervix. The combined effect makes fertilization almost impossible.

Since Depo-Provera provides near-immediate and long-lasting protection, it is typically considered a safe and relatively risk-free way to prevent pregnancy. However, its convenience is not entirely without danger. Like most hormone-altering drugs, Depo-Provera is associated with a litany of side effects. These can include:

  1. Abdominal pain;
  2. Bloating or weight change;

The active ingredient in Depo-Provera is medroxyprogesterone acetate.

Researchers believe that regular exposure to medroxyprogesterone acetate is associated with a higher-than-average risk for meningioma. Meningiomas are a type of tumor that forms in the meninges, a series of protective membranes around the brain and spinal cord. Most meningiomas are benign, meaning that they are not cancerous, but they can still cause serious neurological problems.

Depo-Provera And Brain Tumors

One of the hardest parts of treating meningiomas is that they can be incredibly difficult to detect.

In the earliest stages of growth, meningiomas are typically asymptomatic, and you might not notice that anything is wrong. However, as the tumor grows, more problems can arise, though they are not always obvious. In fact, they are often vague, easily attributable to other, less-serious conditions.

Some of the most common signs of a meningeal tumor include:

  1. Persistent headaches;
  2. Vision problems;
  3. Numbness in the limbs; and
  4. Minor cognitive changes, like a depressed mood or unusual anxiety.
